Effect of adiponectin on the migration of human immune cells. The adipocytokine adiponectin induced the migratory activity of human neutrophil
granulocytes, but did not have any effect on the locomotion of CD8 T lymphocytes.
We mixed 2.5 × 105 neutrophils or T cells with 150 μl of a buffered collagen solution (pH 7.4; 1.67 mg/ml
collagen type I) in the presence of 1 μg/ml adiponectin. The buffered collagen suspension
was filled into self-constructed chambers and allowed to polymerize for 20 min at
37°C in a 5% CO2 humidified atmosphere [9]. We recorded the locomotor activity of the cells by time-lapse videomicroscopy and
subsequently analyzed the migratory behaviour by computer-assisted cell tracking.
The figure shows mean values of three independent experiments (90 cells were analyzed).
